Two men, aged 38 and 44, were charged, with the 38-year-old allegedly repackaging cigarettes and the 44-year-old handling deliveries.
A 41-year-old man was detained after more cigarettes were discovered in a truck and at his home.
Court proceedings are ongoing for the older suspects, while investigations continue for the 41-year-old.
Under Singapore law, smuggling carries fines up to 40 times the evaded amount, up to six years in prison, and vehicle forfeiture.
A separate case involved a 38-year-old Malaysian man arrested at Woodlands Checkpoint on November 1 for smuggling over 250 cartons concealed in a vehicle.
Both cases are under investigation by Singapore Customs.
